New cerebellar ataxia gene identified in dogs

Researchers at the University of Helsinki and the Folkh?lsan Research Center, Finland, have identified the genetic cause of early-onset progressive cerebellar degeneration the Finnish Hound dog breed. The study, led by Professor Hannes Lohi, revealed a new disease mechanism in cerebellar degeneration. A mutation was identified in the SEL1L gene, which has no previous link to inherited cerebellar ataxias.

This gene find is the first in canine early-onset cerebellar degeneration, and has enabled the development of a genetic test to help eradicate the disease from the breed. At the same time, SEL1L represents a novel candidate gene in human early-onset degenerative ataxias.

The research was published in the scientific journal PLoS Genetics on June 14, 2012.

Inherited ataxias affect both humans and animals. In humans, the hereditary ataxias are a heterogeneous disease group, characterized by cerebellar degeneration and dysfunction. The cerebellum is a part of the brain that is involved in coordination of movement. Degeneration of the cerebellar structures causes ataxia, which is a neurological sign of defective motor coordination that can affect gait, balance, speech and gaze. Approximately 20 known disease-causing genes have been identified in both autosomal recessive and dominant ataxias in humans but the genetic background of canine cerebellar ataxias has remained largely unknown.

The clinical signs of Finnish Hound cerebellar ataxia are present by the age of two months. The affected puppies have difficulty in controlling their leg movements and keeping their balance. The disease progresses rapidly, and in the end eating becomes impossible because of uncontrolled head movements. There is no cure for the disease and affected puppies have to be euthanized.

The research conducted by Professor Lohi and co-workers revealed marked neuronal loss in the cerebellar cortex of affected Finnish Hound puppies. By comparing the genomes of affected and healthy dogs, the cause of the disease was pinpointed to a single nucleotide change in the SEL1L gene. The nucleotide alteration causes an amino acid change in the encoded SEL1L protein.

"The identified ataxia gene is the first early-onset degenerative cerebellar ataxia gene that has been identified in dogs", says professor Hannes Lohi. In addition to Finnish Hounds, cerebellar degeneration has been identified in several other dog breeds.

"The SEL1L gene has not been previously connected to cerebellar ataxias in any species and it represents a novel candidate gene for human early-onset ataxias", Lohi continues. "In fact, we have already screened a small cohort of human patients for possible disease-causing SEL1L mutations".

The gene identification in Finnish Hound dogs reveals a new disease mechanism, which will hopefully shed more light in to the pathogenesis of neuronal degeneration. The amino acid change in SEL1L hits an evolutionary conserved functional domain, and is very likely to affect the normal function of the protein. SEL1L functions in quality control of newly synthesized proteins, in a cell organelle referred to as the endoplasmic reticulum. A failure in the quality control system causes endoplasmic reticulum stress and eventually cell death.

"It is very interesting that although the SEL1L gene is expressed in several different tissues, we only saw pathological changes in the cerebellar cortex of affected dogs. It is known that certain neurons in the cerebellar cortex seem to be very sensitive to endoplasmic reticulum stress, which might offer some explanation to the cerebellum-restricted cell loss", tells M.Sc Kaisa Ky?stil?, the first author of the article. "We were also able to measure increased endoplasmic reticulum stress in the affected puppies' cerebellar cortex, which gives more support to SEL1L as the causative gene in Finnish Hound ataxia", Ky?stil? adds.

Finnish Hound ataxia is a single gene defect, inherited in an autosomal recessive manner. According to the research, 10% of Finnish Hounds carries the recessive ataxia mutation in their genome. A genetic test has been developed for breeding purposes. The test has enabled to identify disease carriers, and when mated with non-carriers, they too can be used in breeding programs. If all carrier dogs were excluded by breeders, the genetic diversity of the breed could be affected. The Finnish Hound ataxia test is offered by a DNA-test laboratory Genoscoper (genoscoper.com), and is the first test for a native Finnish breed developed by the research group.

Chinese crew passes first space docking test

China performed its first-ever space docking involving a piloted spacecraft on Monday, joining the ranks of history's two greatest spacefaring nations, the United States and Russia.

China's Shenzhou 9 space capsule ? which launched Saturday carrying three astronauts, including the country's first female spaceflier ? linked up automatically with the unmanned Tiangong 1 space lab just after 2 p.m. Monday Beijing time (2 a.m. ET), according to CCTV.

The only other countries to pull off an orbital docking with a manned spacecraft are the United States and then-Soviet Russia, which first did so in 1966 and 1969, respectively.

Shenzhou 9 was to dock with Tiangong 1 twice. The plan called for the first hookup to be conducted in automated mode, following the pattern set last November during an all-robotic docking between Tiangong and an unmanned Shenzhou 8 craft. At some point, the two spacecraft will separate, and the three taikonauts, as China's astronauts are known, will perform the second docking under manual control.

Forty-six-year-old Jing Haipeng leads the taikonaut crew, which also includes Liu Wang, 42, and 33-year-old Liu Yang, China's first female astronaut. All are members of the Communist Party of China and former pilots with the People's Liberation Army; Jing flew on China's last manned spaceflight, which took place in 2008.

Two of the taikonauts will live aboard Tiangong 1 during the 13-day mission, while one will stay aboard Shenzhou 9 at all times in case of emergency.

Shenzhou 9's flight is considered a key step in China's plan to build a permanently staffed space station in Earth orbit. The nation hopes to have a 60-ton station and up and running by 2020. (For comparison, the International Space Station weighs about 430 tons.)

Shenzhou 9's mission is China's fourth manned spaceflight, following taikonaut launches in 2003, 2005 and 2008. Another manned mission to Tiangong 1 ? which launched to orbit in September 2011 ? could come later this year, Chinese officials have said.

Owning a home is part of the American Dream. Upon reaching that point in life when you are ready to make the big buy, it is important to know how to go about it to avoid making costly mistakes. Expect that the home buying process will be exciting and stressful at the same time, but will be worth it in the end if you make the right moves. Here's a quick guide to help you look at the bigger picture before searching for new homes. Most first time homebuyers often find themselves caught in the whirlwind of finding the perfect house. While the features and fixtures of the structure itself are important, it should not be your immediate priority. It makes more sense to look at the bigger picture before scrutinizing aspects like the number of bedrooms and bathrooms, garage space, and kitchen size.

Location is everything when it comes to new homes for sale. After all, what good are all your dream features if thehouse is in the middle of nowhere? Carefully choosing the community is the first step to a worthwhile investment. Be a smart buyer and keep the following pointers in mind.

City versus the Suburb

Decide whether you want a property in the city or one in the immediately surrounding area. It is the first major decision you have to make and your choice dictates everything else that follows. Think about your budget and the lifestyle you wish to live. Do you want to be close to where the action is all the time? Do you prefer something more laidback but with easy access to a cosmopolitan lifestyle? Answering these questions will help you decide which area to focus on when searching for new homes.

Where you choose to livewillnarrow down your choices in terms of the type of new homes for sale. For example, the city might have more options for condominiums than detached houses, and vice versa for the metro area. This aspect also lays out your options for schools, work, and entertainment.

Economic Stability

General rule of thumb is to buy a home where the economy is flourishing or on the rise. Properties in such locations are good investments because prices are competitive and resale value is high. Thriving cities are also usually host to big industries. This is ideal especially if you seek new job opportunities or career growth to provide for a growing family.

New homes for sale in economically stable locations normally have bigger business districts offering you more choices for entertainment and recreation. Major cities may also be home to bigger school districts providing education across all levels. This is an important inquiry especially if you are interested in a master's or doctoral degree, or want to send your kids to a good school.

Government Services

The last significant concern when choosing a location for new homes is local government services. What's available in terms of hospitals? Does the place have reliable police and fire department services? Are there parks and athletic facilities you can use? These are important aspects most homebuyers tend to overlook. Be a smart buyer and go over all these related aspects before looking into properties one-by-one.

CHICAGO (AP) ? With global anxiety rising, President Barack Obama is searching for bolder, swifter signals from Europe that it will contain its financial mess and keep it from torpedoing the U.S. economy and his re-election chances along with it.

Yet as he prepares for summit talks beginning Monday in Mexico with the other world leaders, Obama is down to the power of persuasion and little else.

A looming, perilous Greek election and Europe's internal politics are controlling the debate.

Given the teetering global economy and the breadth of leaders about to gather in the coastal resort of Los Cabos, the Group of 20 summit meeting carries the weight of expectations it is not likely to meet. Most of its members are not part of Europe and they have no power to drive how the continent manages its crisis, although do they come looking for signs of progress and urgency.

That clearly is the case for Obama, locked in a tight election that may be decided singlehandedly by whether U.S. job growth sinks or climbs over the next five months.

While economic challenge will dominate the summit, the agenda runs deeper.

In talks on the sidelines, Obama will confront the bloodshed in Syria and the nuclear threat in Iran. He will meet with Vladimir Putin, who has returned to the presidency of Russia. Their talks will be scrutinized, given tense U.S.-Russian political relations and deep divisions over Syria.

Obama was to head to Mexico on Sunday night after a weekend with his family in their hometown of Chicago. The summit runs Monday and Tuesday.

Europe's entangled financial crisis, from debt woes in Greece to banking trouble in Spain and high unemployment all around, has become the single biggest threat to the U.S. economic recovery. The signs of worry are clear at the White House and in the words of Obama, who can draw a straight line from the fate of Europe's economic strength to his chances of a second term.

Obama is prodding European leaders to give world markets some confidence, and fast.

"Obviously, this matters to us because Europe is our largest economic trading partner," Obama said. "If there's less demand for our products in places like Paris or Madrid, it could mean less business for manufacturers in places like Pittsburgh or Milwaukee. The good news is there is a path out of this challenge. These decisions are fundamentally in the hands of Europe's leaders."

He wants to emerge from Mexico with signs that the European players at the table, led by Germany, are moving on their own agenda. That means pursuing a banking union to match the monetary union linking the eurozone, taking steps to keep borrowing costs down in the weakest nations and injecting life into economies with growth plans involving public money.

Or in short, as Treasury Undersecretary Lael Brainard put it, the focus in Mexico will be "ensuring our European partners are escalating their response" to stabilizing a dicey situation.

"The stakes are high for all of us," she said.

The agenda threatens to be upended by the outcome of Sunday's election in Greece.

In choosing a new government for their debt-drowning nation, Greek voters are deciding whether to stick with the deeply unpopular austerity terms of an international bailout package or reject them. If they do the latter, that could lead Greece to default and get booted out of the eurozone, which could cause panic and destabilize the world's financial system.

Here, too, Obama has tried to hold sway from abroad.

"It is in everybody's interest for Greece to remain in the eurozone while respecting its commitment to reform," he said. "We recognize the sacrifices that the Greek people have made. ... But the Greek people also need to recognize that their hardships will likely be worse if they choose to exit from the eurozone."

Of the G-20 collection of emerging and economic giants, only Germany, Italy and France are in the 17-nation eurozone at the center of the crisis. European leaders point to a different summit of their own, to be held in Brussels at the end of June, as the more appropriate time to watch for action on a crisis response plan.

Given that dynamic, Obama officials have worked to keep expectations in check.

"Los Cabos will not be the final word on the eurozone," said Mike Froman, deputy national security adviser for international economic affairs.

Even beyond Europe, from Asia to the Americas, the economy is softening and fears are soaring.

The G-20 is meant to serve as the premier voice of economic coordination, representing not just traditional powers but emerging economies. Its members are Argentina, Australia, Brazil, Britain, Canada, China, France, Germany, India, Indonesia, Italy, Japan, Mexico, South Korea, Russia, Saudi Arabia, South Africa, Turkey, the U.S. and the European Union.

Obama's advisers do see the G-20 as a chance for the European nations to add some definition to their plans and show how they will put aside sovereign concerns to avoid calamity.

That pressure can help, said Matthew Goodman, who formerly oversaw the G-20 planning for Obama as director of international economics on the National Security Council staff.

"If you have to go into a meeting with peers and explain yourself, it gives the Europeans some ammunition when they have to try to sell these changes domestically," said Goodman, a specialist in political economy at the Center for Strategic and International Studies. "They can say, 'This is what the world expects.'"
